at the laboratory from the Metropolitan Police, Battersea:

PURPOSE :
I have been asked to examine the above listed items using
the technique of DNA amplification or Polymerase Chain Reaction
( PCR ) in order to determine from whom the bloodstaining
from the lining of the jacket ( AS 12 - areas 1 and 2 )
could have originated.
EXPLANATION OF DNA AMPLIFICATION :
DNA ( Deoxyribonucleic Acid ) is a substance which occurs
in most cells of the body. The chemical structure of DNA
determines the expression of inherited characteristics and
the variation between individuals is, therefore, reflected
in the chemical structure of their DNA.
DNA amplification, otherwise known as Polymerase Chain Reaction
( PCR ) like traditional DNA profiling, is a technique which
analyses variation in the chemical structure of the DNA
between individuals. The technique of DNA amplification
involves targeting specific areas of DNA known to exhibit
variability in their structure, The process then involves
copying these areas many times. This results in the initial
quantity of targeted DNA being greatly increased.
The increased sensitivity resulting from this technique
may be applied to biological samples, such as blood, semen,
saliva, hair roots and bone, where the DNA is of insufficient
quantity or unsuitable quality for traditional DNA profiling
techniques.
EXAMINATION OF ITEMS :
The following analyses have been carried out with the aid
of scientific support staff at the laboratory. The technique
of DNA amplification has been applied to the area of DNA
known as the Human Leucocyte Antigen ( HLA ) DQa locus.
REFERENCE CONTROL SAMPLES :
The HLA DQa region has been successfully amplified and typed
for the blood sample ( MRC 9 - Alison Jane SHAUGHNESSY )
and the root material from some of the head hairs for each
of the samples ( MMS/2511/1 - Michelle Ann TAYLOR and JBH/78/1
- Lisa Jane TAYLOR ).
BLOODSTAINING FROM JACKET : - ( AS 12 - areas 1 and 2 )
Each of the areas of bloodstaining from the lining of the
left sleeve of Jacket have been analysed.
The results of the DNA amplification and typing of the HLA
DQa region for each of the areas of bloodstaining from the
jacket ( AS 12 ) together with the results obtained from
the reference control samples ( MRC 9, MMS/2511/1 ) and
JBH/78/1 ) are presented in a table at the end of this statement.
DISCUSSION :
The head hair samples ( MMS/2511/1 - Michelle Ann TAYLOR
and JBH/78/1 Lisa Jane TAYLOR ) belong to the same HLA DQa
type. The results obtained from the samples of bloodstaining
from the lining of the left sleeve of the jacket ( AS 12
- areas 1 and 2 ) are such that in each instance the blood
could have originated from either Michelle Ann TAYLOR or
Lisa Jane TAYLOR. This blood could not have originated from
Alison Jane SHAUGHNESSY.
CONCLUSIONS :
The above findings are consistent with the bloodstaining
from the lining of the left sleeve of the jacket ( AS 12
) having originated from either Michelle Ann TAYLOR or Lisa
Jane TAYLOR.

Note 1 :
The method used to determine the HLA DQa genotype employs
the commercially produced Cetus Amplitype TM HLA DQa Forensic
DNA Amplification and Typing Kit.
